What is a Prokaryote?

Prokaryote: A prokaryote is a single-celled organism that lacks a nucleus and membrane-bound organelles. Its genetic material is found in a single, circular DNA molecule located in the nucleoid region of the cell.

Prokaryotes are among the oldest and simplest forms of life on Earth. Despite their structural simplicity, they are extraordinarily successful organisms , bacteria and archaea are found in almost every environment imaginable. Understanding their structure is essential for appreciating how life can function with minimal complexity.

What is a Prokaryote?

Prokaryote Cell Structure: Essential Components

A prokaryotic cell has four essential structural components: a cell wall, a plasma membrane, cytoplasm, and naked DNA in a nucleoid region, plus 70S ribosomes for protein synthesis. Each plays a distinct and vital role.

1. Cell Wall

Cell wall: The cell wall is a rigid, protective layer located outside the plasma membrane. In prokaryotes, it is composed of peptidoglycan , a polymer of sugars and amino acids forming a rigid mesh , providing structural support and protection.

  • The thick peptidoglycan layer gives the cell its shape and protects it from environmental stresses such as osmotic pressure.
Note

Gram-positive bacteria stain purple during Gram staining because their thick peptidoglycan layer retains the crystal violet dye. This is a medically important distinction.

2. Plasma Membrane

Plasma membrane: The plasma membrane is a thin, flexible phospholipid bilayer with embedded proteins that separates the cell's interior from its surroundings.

  • It controls the transport of nutrients and waste.
  • It maintains homeostasis by regulating ion concentrations.
  • In prokaryotes, it is the site of ATP production during cellular respiration , a function carried out by mitochondria in eukaryotes.

3. Cytoplasm

Cytoplasm: The cytoplasm is the gel-like substance filling the interior of the cell, containing enzymes, ribosomes, and the nucleoid region. It is the site of most metabolic processes.

  • Unlike eukaryotes, prokaryotic cytoplasm has no membrane-bound compartments , all reactions occur in a single open space.
Warning

Avoid confusing cytoplasm with cytosol. The cytoplasm includes the cytosol (the fluid portion) plus all suspended structures such as ribosomes and DNA.

4. Naked DNA (Nucleoid Region)

Naked DNA: Naked DNA refers to DNA that is not enclosed within a nucleus or bound to histone proteins. In prokaryotes, it exists as a single, circular molecule free in the cytoplasm.

Nucleoid: The nucleoid is the region of the cytoplasm where the circular prokaryotic DNA molecule is located. It is less dense than the surrounding cytoplasm and is visible under an electron microscope.

  • Many prokaryotes also carry smaller DNA molecules called plasmids, which often encode advantageous traits such as antibiotic resistance.

5. 70S Ribosomes

70S Ribosomes: 70S ribosomes are the smaller ribosomes found in prokaryotic cells. They are responsible for protein synthesis, translating mRNA into functional proteins. The "S" refers to Svedberg units , a measure of sedimentation rate during centrifugation.

  • 70S ribosomes are scattered throughout the cytoplasm and are highly efficient despite their small size.
Exam Technique

If asked to distinguish prokaryotic from eukaryotic cells, ribosome size is a key mark-scoring feature: prokaryotes have 70S ribosomes, eukaryotes have 80S ribosomes. This difference is also clinically important , many antibiotics specifically target 70S ribosomes without harming human cells.
